Hoechst 33258，也稱bisBenzimide H 33258或HOE 33258，是一種可以穿透細胞膜的藍色熒光染料，對細胞的毒性較低。Hoechst 33258染色常用于細胞凋亡檢測，染色后用熒光顯微鏡觀察或流式細胞儀檢測。Hoechst 33258也常用于普通的細胞核染色，或常規(guī)的DNA染色。Hoechst 33258的*激發(fā)波長為346nm，*發(fā)射波長為460nm；Hoechst 33258和雙鏈DNA結(jié)合后，*激發(fā)波長為352nm，*發(fā)射波長為461nm。本Hoechst 33258染色液可直接用于固定細胞或組織的細胞核染色，也可直接用于活細胞或組織的細胞核染色。
使用說明： 使用前用生理鹽水或PBS將Hoechst 33258染色液稀釋100倍，即為工作液。
1. 對于固定的細胞或組織：
a. 對于細胞或組織樣品，固定后，適當洗滌去除固定劑。隨后如果需要進行免疫熒光染色，則先進行免疫熒光染色，染色完畢后再按后續(xù)步驟進行Hoechst 33258染色。如果不需要進行其它染色，則直接進行后續(xù)的Hoechst 33258染色。
b. 對于貼壁細胞或組織切片，加入少量Hoechst 33258工作液，覆蓋住樣品即可；對于懸浮細胞，至少加入待染色樣品3倍體積的工作液，混勻。室溫放置3-5分鐘。
c. 吸除Hoechst 33258染色液，用TBST、PBS或生理鹽水洗滌2-3次，每次3-5分鐘。
d. 直接在熒光顯微鏡下觀察或封片后熒光顯微鏡下觀察。細胞發(fā)生凋亡時，會看到凋亡細胞的細胞核呈致密濃染，或呈碎塊狀致密濃染。
2. 對于活細胞或組織：
a. 加入適當量Hoechst 33258工作液，必須充分覆蓋住待染色的樣品，通常對于六孔板一個孔需加入1mL工作液，對于96孔板一個孔需加入100μL工作液。
b. 在適宜于細胞培養(yǎng)的溫度下培養(yǎng)20-30分鐘。棄染色液，用PBS或培養(yǎng)液洗滌2-3次即可進行熒光檢測。

熒光染料都存在淬滅的問題，為減緩熒光淬滅可以使用抗熒光衰減封片劑。建議染色后盡量當天完成檢測，活細胞或組織染色后應立即觀察。
