蘇木素是一種堿性染料，同媒染劑（常用的是三價(jià)的鐵或鋁的鹽）一起使用，能夠使細(xì)胞核染色。蘇木素和氧化蘇木素都沒(méi)有染色能力，但與媒染劑結(jié)合形成色淀（lake），這種色淀具有染色能力。核、染色體、著絲粒、中心體、線粒體、髓鞘等可被染成藍(lán)色以至黑色。與伊紅合用的蘇木素-伊紅雙重染色法是最基本的一般組織的染色法，蘇木素染色原理如下：
蘇木素和伊紅在組織學(xué)上被經(jīng)常使用，碘液不同，這是一種*染色街。其他常用的含有蘇木素的染色劑有磷鎢酸蘇木素染色劑，也就是磷鎢酸與蘇木素的混合物。蘇木素染色劑經(jīng)常為組織的研究使用。明礬和三價(jià)鐵鹽常常被用作媒染劑，展示核和細(xì)胞質(zhì)結(jié)構(gòu)。這些媒染劑的原理都是形成媒染劑－染料－組織復(fù)合體，從而顯示顏色。
蘇木素染色內(nèi)質(zhì)呈淡藍(lán)黑色，外質(zhì)收縮，剩下部分不著色或色澤更淺。圓形細(xì)胞核呈藍(lán)黑色。居中的核仁呈黑色小圓點(diǎn)。核膜較薄，其內(nèi)側(cè)緣均勻，整齊地排列著一層細(xì)小的染色質(zhì)粒。核仁與核膜之間可見(jiàn)到核網(wǎng)。被吞噬的紅細(xì)胞呈藍(lán)黑色采用蘇木素染色液進(jìn)行免疫染色后的復(fù)染，操作步驟簡(jiǎn)便，操作時(shí)間短?？梢圆僮鞑襟E簡(jiǎn)便，操作時(shí)間短或培養(yǎng)細(xì)胞的染色，或與免疫組織化學(xué)、免疫細(xì)胞化學(xué)、免疫熒光、原位雜交等配合使用。使用蘇木素染色液染色后還可以進(jìn)行免疫染色或其它染料的復(fù)染。蘇
蘇木素染色原理很簡(jiǎn)單，蘇木素染色法也是常用的一種染色方法。木素染色后的細(xì)胞核呈現(xiàn)藍(lán)色，細(xì)胞漿呈現(xiàn)粉紅色或紅色。染色液也可以重復(fù)使用，直至認(rèn)為效果不佳時(shí)再換用新的染色液。
